Mercurimetric determination of chloride in biologic fluids using 2-(8-hydroxyquinolyl-5-azo)- benzoic acid (QBA).

ITRATION of the chloride ion with Hg(II), using s-diphenylcarbazone as an indicator has been discussed by Laitinen (1). This is a one-color indicator which at pH 3 is colorless in the unmetallized form and forms a violet Hg(II) complex. The violet color is seen at the end point of the titration. Schales and Schales (2) used s-diphenylcarbazone as the indicator for the mercurinletric titration of the chloride ion in body fluids; a variant of this now widely used and accepted method is employed here as the reference method. The dyestuff, 2(8-hydroxyquinolyl-5-azo) -benzoic acid (QBA), which had been found to be suitable as a mercurimetric indicator by Sivanova (3), was evaluated for chloride determination in human blood serum. The data presented here show QBA to be completely suitable as an indicator for this determination at about pH 3 and a satisfactory alternative for s-diphenylcarbazone. The nonmetallized form of QBA is orange in a weakly acidic solution. The Hg(II) derivative is rose-red. The Hg(II) cation reversibly displaces the proton from the 8-hydroxy group on the quinoline nucleus of the QBA. The participation of the proton in the equilibrium makes this reaction dependent upon tile pTT of the svsten#{238}.
